Synthetic biology is a newly developed interdisciplinary research field combining molecular biology with engineering design. This field considers that living systems are programmable at the genetic level. Synthetic biology makes the applications of systematic design approaches to construct novel cells or biological systems with human-defined functions possible. One of the key principles of synthetic biology is the Design-Build-Test-Learn (DBTL) cycle, which aims to fulfill specific design criteria for a synthetic biology application. The design phase and build phase mainly focus on host organism selection, modeling, simulation, and construct the organisms based on the planned design. The test phase covers the in-depth testing of the organisms. The results are analyzed at the final “Learn” phase. The DBTL cycle is an increasingly adopted metabolic engineering framework. It is facilitated by exponentially improving capabilities in synthetic biology.
With the increasing digitization of biological data, there is a need to develop enabling technologies and platforms that allow algorithm-driven biological design. Computer-aided approaches can be combined with DNA assembly for design and build. A variety of biological data, primarily multi-omics data, are usually measured and assessed in the test phase. These datasets can be used as part of the learn phase. This systematic engineering approach for the construction of living systems allows the application of synthesis-based foundational tool platforms to many different application areas. This is also the difference between synthetic biology and more traditional biotechnology or metabolic engineering.
