We primarily use isothermal assembly (also known as Gibson assembly), which combines exonuclease, DNA polymerase, and DNA ligase in a single reaction at 50°C. This method requires 30-40 bp overlapping regions between fragments and provides highly accurate assembly of multiple DNA fragments.
We offer a wide range of modifications including E1/E3 gene deletions for replication-defective vectors, fiber protein modifications (RGD, Ad5/F35 chimeras) for altered tropism, and transgene insertion in E1 or E3 regions. Gutless adenovirus constructs are also available upon request.
Every clone undergoes comprehensive quality control including complete genome sequencing by Sanger sequencing, restriction enzyme analysis for pattern verification, and clone stability testing. For constructs larger than 5 kb, NGS verification is performed to ensure 100% sequence accuracy.
