Gene: PCDHG@
Official Full Name: protocadherin gamma clusterprovided by HGNC
Gene Summary: The protocadherin gamma gene cluster is one of three related clusters tandemly linked on chromosome five. The clusters have an immunoglobulin-like organization, suggesting that a novel mechanism may be involved in their regulation and expression. The gamma cluster includes 22 genes divided into 3 subfamilies, all part of the cadherin superfamily. Subfamily A contains 12 genes, subfamily B contains 7 genes and 2 pseudogenes, and the more distantly related subfamily C contains 3 genes. In addition, ORNT2 and TAF2F are included in the cluster due to their localization between protocadherin clusters beta and gamma on chromosome five. The protocadherin genes are organized in a tandem array of 22 large, variable region exons followed by a constant region, which contains 3 exons shared by all protocadherin genes in the cluster. Each variable region exon encodes an extracellular domain comprised of 6 cadherin ectodomains and a transmembrane region. The constant region exons encode a common cytoplasmic tail. These neural adhesion proteins most likely play a critical role in the establishment and function of specific cell-cell connections in the brain. Alternative splicing has been described for the gamma cluster genes. [provided by RefSeq, Jul 2008]

PCDHG@ Gene Knockout Cell Lines are precisely engineered cellular systems that lack functional PCDHG (protocadherin gamma) genes, enabling researchers to study the biological roles and implications of these genes in both physiological and pathological conditions. These cell lines are constructed using state-of-the-art CRISPR/Cas9 gene-editing technology, ensuring high efficiency and specificity in gene disruption. The knockout of PCDHG facilitates functional analyses, paving the way for insights into cell adhesion, signaling pathways, and neural development, as protocadherins are integral to the formation and function of synapses in the central nervous system.
The primary mechanism of action revolves around the targeted modification of the PCDHG gene locus, resulting in a complete loss of gene expression. This allows for investigations into gene function, cellular response mechanisms, and potential treatments for diseases linked to PCDHG abnormalities, such as neurodevelopmental disorders. In research contexts, these cell lines are invaluable tools for elucidating the roles of protocadherins in cellular interactions and their broader implications in developmental biology and neurobiology.
One of the major advantages of the PCDHG@ Gene Knockout Cell Lines is their reproducibility and reliability compared to other models that rely on transient or less precise gene silencing methods. Unlike RNA interference techniques, which may result in partial or off-target effects, these knockout lines yield consistent results essential for robust scientific conclusions. This specificity positions them as a superior alternative, providing enhanced experimental control for researchers focused on understanding the complex functions of neuronal cell adhesion molecules.
For researchers and clinicians dedicated to uncovering the underlying mechanisms of neural development and disorders, PCDHG@ Gene Knockout Cell Lines offer remarkable value.
