Description Staphylococcus aureus Cas9 is expressed as a C-terminal 6×His-tagged fusion in E. coli. It is a programmable DNA endonuclease guided by a sgRNA. Staphylococcus aureus Cas9 can catalyze site-specific cleavage of double-stranded DNA. A ~100 nucleotide gRNA complementary to the target site and a 5′ NNGRRT 3′ (N = any, R = A or G) protospacer adjacent motif (PAM) on the DNA strand opposite the target sequence are required for targeting.
