We deliver custom mammalian cell lines with site-specific CRISPR/Cas9 integration of your multi-gene pathway cassette for stable, long-term expression.
Random integration can cause position effects, silencing, and inconsistent expression. Our CRISPR-based approach places your entire pathway into a defined genomic locus, ensuring reproducible and stable output across passages.
From construct design to validated cell banks, our integrated workflow is tailored to your pathway’s size, complexity, and intended application.
We engineer your multi-gene pathway into a donor template with homology arms targeting your chosen genomic locus. gRNAs are optimized.
Following transfection, we apply antibiotic selection and perform single-cell cloning to isolate stable integrants. Each clone is verified by junction PCR and sequencing, then functionally assessed for pathway activity.
Site-specific integration reduces line-to-line expression variability, supporting consistent results across experiments and scale-up campaigns.
Choose from basic integration confirmation (PCR + sequencing) to full functional pathway characterization, depending on your project goals.
A dedicated scientific contact guides you from design to final report, with milestone updates at each step.
All deliverables are provided with detailed records to support your downstream applications.
Each project is scoped individually based on your target cell type, pathway cassette, and validation needs. Typical parameters are listed below.
| Parameter | Typical project scope |
|---|---|
| Target cell line | Mammalian cells (HEK293, CHO, HeLa, U2OS, etc.); other lines assessed on request |
| Pathway cassette size | Scoped per project after consultation |
| gRNA design approach | In silico design with off-target scoring; two gRNAs per target site recommended |
| Doner template format | Scoped per project after consultation |
| Integration verification | Junction PCR (5′ and 3′ junctions) + Sanger sequencing; copy number analysis optional |
| Functional validation | Metabolite production assay (LC-MS or colorimetric) or reporter activity; custom assays scoped per project |
| Clonal output | Scoped per project after consultation |
Our five-stage workflow keeps your project on track from concept to delivered cell line.
We review your pathway sequence, select a target genomic locus, design homology arms and gRNAs, and run off-target predictions. A design report is shared for your approval before proceeding.
The multi-gene donor cassette is assembled into a plasmid backbone with homology arms and a selection marker. gRNA expression vectors are constructed separately. All constructs are verified by restriction digest and Sanger sequencing.
Mammalian cells are transfected with Cas9, gRNA, and donor plasmids. After 48–72 hours, antibiotic selection is applied to enrich for integrants. The polyclonal pool is cryopreserved as an intermediate deliverable.
The polyclonal pool is plated at limiting dilution to isolate single cells. Clones are expanded over a timeline confirmed during project scoping, and genomic DNA is extracted for integration screening.
Each candidate clone is tested by junction PCR and Sanger sequencing to confirm correct integration. Positive clones undergo functional pathway assay. Final cell banks are cryopreserved, and a project report including methods, QC, and datasets is delivered.
Stable integration of multi-gene pathways enables a wide range of bioproduction and cell engineering projects.
Engineer cells to express biosynthetic pathways for small molecules, antibodies, or therapeutic proteins with stable, defined genetic context.
Integrate inducible or constitutive reporter cassettes, multi-enzyme cascades, or sensing circuits for cellular engineering and assay development.
Create cell models expressing disease-relevant pathway variants or entire metabolic circuits for drug screening and target discovery.
Straightforward answers to help you plan your project.
We routinely work with HEK293, CHO, HeLa, and U2OS cells.Other adherent mammalian lines can be accommodated after a feasibility assessment during project scoping.
Validation is scoped per project. For metabolic pathways, we measure target metabolite levels (e.g., by LC-MS or colorimetric assay). For reporter pathways, we quantify fluorescence or luminescence. The assay used is agreed upon at the design stage.
We design two gRNAs per target site and may test multiple homology arm lengths if needed. The use of small-molecule HDR enhancers can also be incorporated as a project option. Efficiency is evaluated during the polyclonal pool step, and alternative strategies are discussed if the initial yield is insufficient.
Single-cassette integration becomes more challenging beyond 10 kb. For larger pathways, we recommend splitting into two separate integration sites or using a bicistronic/multi-promoter design within the size limit. We assess feasibility on a per-project basis.
