Our Approach

Precision Gene Repression in C. glutamicum

We combine computational sgRNA design with robust dCas9 delivery to achieve efficient, specific transcriptional silencing in your target strain.

Rational sgRNA Design

Bioinformatic design of single guide RNAs against target gene promoters or coding regions, optimized for C. glutamicum dCas9 activity and minimal off-target binding.

  • Promoter or coding strand targeting
  • Off-target in silico analysis

dCas9 Vector Construction

Cloning of validated sgRNAs into a CRISPRi plasmid carrying a catalytically dead Cas9 (dCas9) under a constitutive or inducible promoter, with selection markers suited for C. glutamicum.

  • Inducible or constitutive dCas9 expression
  • E. coli–C. glutamicum shuttle vector

Transformation & Selection

Electroporation-mediated transformation into C. glutamicum, followed by antibiotic selection and colony PCR verification of plasmid maintenance.

Validation

Measurement of target gene repression by qRT-PCR and, where applicable, by growth phenotype or metabolite titer to confirm functional silencing.

Multiplex Capability

Design and assembly of multiple sgRNAs in a single vector for simultaneous repression of several genes, enabling pathway-wide engineering.

Service Scope

Project Customization Parameters

Each CRISPRi project is scoped individually based on your target genes, desired repression level, and downstream application. Key customization parameters include:

Parameter Typical project scope
Target Gene(s) Single or up to multiple genes; promoter or coding strand target site selected during design
sgRNA Design Strategy Bioinformatic design with off-target checking; number of sgRNAs per gene as scoped
dCas9 Expression System Constitutive or inducible promoter (e.g., Ptac, Ptrc); dCas9-mediated steric repression is the standard bacterial mechanism
Vector Selection Marker Kanamycin, chloramphenicol, or spectinomycin resistance; chosen per host strain
Validation Method QRT-PCR for gene expression; optional phenotypic or metabolite assay depending on project goals
Multiplexing Multiple sgRNAs in a single vector; array or polycistronic design evaluated during consultation
Workflow

How It Works

Our streamlined CRISPRi service proceeds through five defined stages, from design to validated strain delivery.

Target Design & sgRNA Synthesis

We design sgRNAs targeting your gene(s) of interest using C. glutamicum-specific algorithms, synthesize oligonucleotides, and clone them into the CRISPRi vector.

dCas9 Vector Assembly

The sgRNA expression cassette and dCas9 are assembled into a shuttle vector. Sequence verification confirms correct cloning before transformation.

Transformation into C. glutamicum

Electrocompetent C. glutamicum cells are transformed with the validated CRISPRi plasmid. Transformed colonies are selected.

Colony Screening & Plasmid Maintenance

Individual colonies are screened by colony PCR and plasmid extraction to confirm the presence of the full CRISPRi construct. Positive clones are expanded.

Repression Validation

Target gene expression is quantified by qRT-PCR relative to a control. If applicable, growth or production phenotypes are assessed. A final report is compiled.

Applications

Common Uses of CRISPRi in C. glutamicum

Our service supports a range of metabolic engineering and research goals in this industrial workhorse.

Pathway Optimization

Reversibly repress competing pathways or enzymes that divert flux away from the target product to redirect carbon flow toward desired products such as amino acids, organic acids.

  • Downregulate side reactions
  • Fine-tune expression levels
  • Iterative design-build-test cycles

Gene Essentiality & Phenotype Screening

Rapidly knock down gene expression to assess function. CRISPRi allows conditional knockdowns to study essential or conditionally essential genes without permanent knockout.

  • Conditional knockdown
  • Phenotypic characterization
  • Target identification

Multiplex Repression for Improved Productivity

Combine multiple sgRNAs in one vector to simultaneously repress several genes, accelerating strain engineering for higher titers or yields.

  • Multiplex targeting
  • Balanced pathway engineering
  • Rapid prototyping
FAQ

Frequently Asked Questions

Common questions about our CRISPRi service for Corynebacterium glutamicum.

  • Project timelines depend on the number of target genes, validation depth, and complexity. Typical projects range from 6 to 10 weeks, including design, construction, transformation, and validation. Your project manager will provide a detailed schedule during the scoping phase.

  • Yes, we routinely design and assemble vectors carrying multiple sgRNAs for multiplex repression. The number of sgRNAs and their arrangement (array or polycistronic) are evaluated during the consultation to support efficient repression of all targets.

  • We perform in silico off-target analysis for each sgRNA against the C. glutamicum genome. Functional off-target repression can be assessed by expression analysis of plausible off-target genes when included in scope; sequencing alone cannot confirm transcriptional specificity.

  • Each project includes a control strain transformed with the empty CRISPRi vector (dCas9 without sgRNA) and, when relevant, a non-targeting sgRNA control. These controls help assess background effects; independent guides or complementation can strengthen attribution of a phenotype to the intended target.
