Gene: IGH
Official Full Name: immunoglobulin heavy locusprovided by HGNC
Gene Summary: Immunoglobulins recognize foreign antigens and initiate immune responses such as phagocytosis and the complement system. Each immunoglobulin molecule consists of two identical heavy chains and two identical light chains. This region represents the germline organization of the heavy chain locus. The locus includes V (variable), D (diversity), J (joining), and C (constant) segments. During B cell development, a recombination event at the DNA level joins a single D segment with a J segment; this partially rearranged D-J gene is then joined to a V segment. The rearranged V-D-J is then transcribed with the IGHM constant region; this transcript encodes a mu heavy chain. Later in development B cells generate V-D-J-Cmu-Cdelta pre-messenger RNA, which is alternatively spliced to encode either a mu or a delta heavy chain. Mature B cells in the lymph nodes undergo switch recombination, so that the V-D-J gene is brought in proximity to one of the IGHG, IGHA, or IGHE genes and each cell expresses either the gamma, alpha, or epsilon heavy chain. Recombination of many different V segments with several J segments provides a wide range of antigen recognition. Additional diversity is attained by junctional diversity, resulting from the random addition of nucleotides by terminal deoxynucleotidyltransferase, and by somatic hypermutation, which occurs during B cell maturation in the spleen and lymph nodes. Due to polymorphism, the numbers of functional V, J, and D genes differ among individuals and some V, D, J, and C segments may be pseudogenes. [provided by RefSeq, Dec 2017]

IGH Gene Knockout Cell Lines are specifically engineered cellular models that have undergone targeted gene disruption to eliminate the expression of the immunoglobulin heavy chain gene (IGH) within a defined cellular context. These cell lines provide a crucial tool for researchers to dissect the roles of IGH in immunological processes, B cell development, and antibody production. By utilizing techniques such as CRISPR/Cas9 gene editing, these cell lines ensure precise gene knockout, thereby allowing scientists to study the functional consequences of IGH absence in various biological assays.
The key mechanism by which IGH gene knockout cell lines operate involves the targeted deletion of the IGH locus, which effectively halts the production of functional immunoglobulin M (IgM) and other antibody isotypes subsequently. This alteration enables researchers to investigate the resulting phenotypic changes in B cells, including differentiation, signaling pathways, and the cellular environment's response to pathogen exposure or other immune challenges.
In clinical and research settings, these knockout cell lines are invaluable for understanding autoimmune diseases, B cell malignancies, and the development of therapeutic antibodies. The ability to screen potential treatments in a controlled environment accelerates the discovery of novel immunotherapeutic agents and aids in vaccine development, as these lines can mimic disease states without the confounding effects of intact IGH function.
Compared to traditional cell lines that express IGH, our IGH Gene Knockout Cell Lines offer significant advantages, including enhanced specificity in research outcomes and reduced variability in experimental results. Researchers can be confident that observed effects are directly attributable to the absence of IGH, facilitating the interpretation of data and leading to more robust conclusions.
