PTEN Knockout Cell Lines

Gene: PTEN

Official Full Name: phosphatase and tensin homologprovided by HGNC

Gene Summary: This gene was identified as a tumor suppressor that is mutated in a large number of cancers at high frequency. The protein encoded by this gene is a phosphatidylinositol-3,4,5-trisphosphate 3-phosphatase. It contains a tensin like domain as well as a catalytic domain similar to that of the dual specificity protein tyrosine phosphatases. Unlike most of the protein tyrosine phosphatases, this protein preferentially dephosphorylates phosphoinositide substrates. It negatively regulates intracellular levels of phosphatidylinositol-3,4,5-trisphosphate in cells and functions as a tumor suppressor by negatively regulating AKT/PKB signaling pathway. The use of a non-canonical (CUG) upstream initiation site produces a longer isoform that initiates translation with a leucine, and is thought to be preferentially associated with the mitochondrial inner membrane. This longer isoform may help regulate energy metabolism in the mitochondria. A pseudogene of this gene is found on chromosome 9. Alternative splicing and the use of multiple translation start codons results in multiple transcript variants encoding different isoforms. [provided by RefSeq, Feb 2015]

Background

PTEN Gene Knockout Cell Lines are specifically engineered cellular models where the PTEN gene, a critical tumor suppressor, has been inactivated. This product serves as an invaluable tool for researchers investigating the role of PTEN in various biological processes, particularly its involvement in cellular signaling pathways that regulate cell growth, proliferation, and survival. By removing PTEN, these cell lines mimic the oncogenic environment found in certain cancers, providing a platform for studying tumor biology.

The primary mechanism by which PTEN knockout impacts cellular function involves the dysregulation of the phosphoinositide 3-kinase (PI3K)/Akt pathway. In the absence of functional PTEN, this pathway is often constitutively activated, leading to increased cell survival and proliferation. Understanding these pathways in detail can unveil potential therapeutic targets for inhibiting tumor growth and progression, making these cell lines indispensable in cancer research and drug development.

The scientific importance of PTEN Gene Knockout Cell Lines lies in their application across various fields including oncology, genetics, and pharmacology. These models facilitate the exploration of cancer therapeutics and offer insights into disease mechanisms, significantly aiding the development of targeted treatments. Additionally, they are essential for validating the efficacy of drug candidates designed to counteract uncontrolled cell proliferation.

Compared to alternative models used in cancer research, PTEN knockout cell lines offer precise manipulation of the PTEN gene, ensuring specificity and relevance in experimental outcomes. This unique feature greatly enhances the ability of researchers to draw meaningful conclusions and leads to a more thorough understanding of PTEN’s tumor suppressive functions.

The value of PTEN Gene Knockout Cell Lines extends to researchers and clinicians who are focused on advancing their understanding of cancer biology and developing novel therapeutic approaches. These cell lines not only streamline experimental workflows but also support groundbreaking discoveries in cancer treatment.
