SIRT1 Knockout Cell Lines

Gene: SIRT1

Official Full Name: sirtuin 1provided by HGNC

Gene Summary: This gene encodes a member of the sirtuin family of proteins, homologs to the yeast Sir2 protein. Members of the sirtuin family are characterized by a sirtuin core domain and grouped into four classes. The functions of human sirtuins have not yet been determined; however, yeast sirtuin proteins are known to regulate epigenetic gene silencing and suppress recombination of rDNA. Studies suggest that the human sirtuins may function as intracellular regulatory proteins with mono-ADP-ribosyltransferase activity. The protein encoded by this gene is included in class I of the sirtuin family. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Dec 2008]

Background

SIRT1 Gene Knockout Cell Lines are genetically engineered cell lines in which the SIRT1 gene has been selectively disrupted, producing a model for studying the various biological roles of SIRT1 within cellular processes. SIRT1, a member of the sirtuin family of proteins, is pivotal in regulating cellular metabolism, stress responses, and aging. By employing CRISPR/Cas9 or other targeted gene-editing technologies, these knockout cell lines allow researchers to elucidate the physiological effects of SIRT1 absence, providing insights into associated pathways involved in metabolic diseases, cancer, and age-related conditions.

In terms of functionality, the SIRT1 knockout cell lines serve as a vital tool for understanding the mechanisms by which SIRT1 influences key cellular processes such as DNA repair, apoptosis, and telomere maintenance. These mechanistic insights are crucial for developing therapeutic strategies that leverage the modulation of SIRT1 activity in the treatment of diseases including diabetes, neurodegenerative disorders, and specific types of cancer.

The scientific importance of these cell lines lies in their ability to facilitate cutting-edge research and therapeutic advancements. In clinical settings, they can be instrumental in drug discovery, allowing researchers to screen potential SIRT1 modulators in a controlled environment. Furthermore, they provide a robust platform for studying the impact of dietary and environmental factors on SIRT1 activity and subsequent cellular outcomes.

What sets SIRT1 Gene Knockout Cell Lines apart from other genetic models is their specificity, allowing for precise targeting of the SIRT1 gene while minimizing off-target effects. This specificity, coupled with validated performance, ensures reliable and reproducible results, making them an indispensable resource for researchers aiming to deepen their understanding of sirtuin biology.

For researchers and clinicians alike, the value of SIRT1 Gene Knockout Cell Lines is evident: they not only enhance the rigor of scientific inquiry but also pave the way for breakthroughs in targeted therapies.
