PKD1 Knockout Cell Lines

Gene: PKD1

Official Full Name: polycystin 1, transient receptor potential channel interactingprovided by HGNC

Gene Summary: This gene encodes a member of the polycystin protein family. The encoded glycoprotein contains a large N-terminal extracellular region, multiple transmembrane domains and a cytoplasmic C-tail. It is an integral membrane protein that functions as a regulator of calcium permeable cation channels and intracellular calcium homoeostasis. It is also involved in cell-cell/matrix interactions and may modulate G-protein-coupled signal-transduction pathways. It plays a role in renal tubular development, and mutations in this gene cause autosomal dominant polycystic kidney disease type 1 (ADPKD1). ADPKD1 is characterized by the growth of fluid-filled cysts that replace normal renal tissue and result in end-stage renal failure. Splice variants encoding different isoforms have been noted for this gene. Also, six pseudogenes, closely linked in a known duplicated region on chromosome 16p, have been described. Background

PKD1 Gene Knockout Cell Lines are genetically engineered cell lines designed to specifically disrupt the expression of the PKD1 gene, which encodes for polycystin-1, a crucial protein involved in kidney function and development. By employing advanced CRISPR-Cas9 gene editing technology, these knockout cell lines facilitate the detailed study of PKD1's role in cellular processes and disease mechanisms, particularly in autosomal dominant polycystic kidney disease (ADPKD).

The primary function of the PKD1 Gene Knockout Cell Lines is to serve as a valuable tool for researchers exploring the pathophysiology of ADPKD and other related conditions. By eliminating PKD1 expression, these cell lines enable scientists to observe alterations in cellular signaling pathways, proliferation rates, and response to various stimuli, thereby elucidating the gene's function and its implication in disease progression. This functionality makes the PKD1 knockout model an indispensable resource for drug discovery and therapeutic intervention studies.

In the scientific community, the importance of these knockout cell lines cannot be overstated. They have broad applications in both research and clinical settings, allowing for high-throughput screening of therapeutic compounds and the validation of new treatment strategies aimed at mitigating the effects of polycystic kidney disease. These cell lines also aid in the understanding of the molecular mechanisms behind kidney cyst formation and progression, thus contributing to improved patient outcomes.

Compared to alternative models such as standard immortalized cell lines or animal models, PKD1 Gene Knockout Cell Lines offer unique advantages. Their use of human-derived cellular systems ensures higher relevance to human physiology and disease, while the knockout nature of the lines allows for direct observation of the absence of PKD1. Additionally, they streamline experiments by providing a consistent and readily available source of cells, which reduces variability often seen with more complex models.

For researchers, clinicians, and biopharmaceutical companies focused on kidney diseases, these genetically modified cell lines represent an essential advancement, offering clearer insights and more effective pathways to explore potential therapies. Our company, dedicated to providing leading-edge biological products, brings a strong expertise in genetic engineering and cell line development, ensuring that researchers have access to reliable and innovative tools that drive discovery in the field of nephrology and beyond.
