PIKFYVE Knockout Cell Lines

Gene: PIKFYVE

Official Full Name: phosphoinositide kinase, FYVE-type zinc finger containingprovided by HGNC

Gene Summary: Phosphorylated derivatives of phosphatidylinositol (PtdIns) regulate cytoskeletal functions, membrane trafficking, and receptor signaling by recruiting protein complexes to cell- and endosomal-membranes. Humans have multiple PtdIns proteins that differ by the degree and position of phosphorylation of the inositol ring. This gene encodes an enzyme (PIKfyve; also known as phosphatidylinositol-3-phosphate 5-kinase type III or PIPKIII) that phosphorylates the D-5 position in PtdIns and phosphatidylinositol-3-phosphate (PtdIns3P) to make PtdIns5P and PtdIns(3,5)biphosphate. The D-5 position also can be phosphorylated by type I PtdIns4P-5-kinases (PIP5Ks) that are encoded by distinct genes and preferentially phosphorylate D-4 phosphorylated PtdIns. In contrast, PIKfyve preferentially phosphorylates D-3 phosphorylated PtdIns. In addition to being a lipid kinase, PIKfyve also has protein kinase activity. PIKfyve regulates endomembrane homeostasis and plays a role in the biogenesis of endosome carrier vesicles from early endosomes. The protein plays a key role in cell entry of ebola virus and SARS-CoV-2 by endocytosis Mutations in this gene cause corneal fleck dystrophy (CFD); an autosomal dominant disorder characterized by numerous small white flecks present in all layers of the corneal stroma. Histologically, these flecks appear to be keratocytes distended with lipid and mucopolysaccharide filled intracytoplasmic vacuoles. [provided by RefSeq, Jul 2021]

Background

PIKFYVE Gene Knockout Cell Lines are genetically engineered cell lines where the PIKFYVE gene has been disrupted, allowing researchers to study the specific biological functions and pathways associated with this crucial phosphoinositide 5-kinase. By providing a definitive model to assess the effects of PIKFYVE deletion, these cell lines enable in-depth investigations into cell signaling pathways and membrane dynamics that are critical for various cellular processes, including endocytosis and autophagy.

The key function of PIKFYVE involves the synthesis of phosphatidylinositol 5-phosphate (PI5P) and phosphatidylinositol 3,5-bisphosphate (PI(3,5)P2), which are vital for maintaining the integrity of membrane trafficking and signaling. These knockout cell lines allow researchers to elucidate the role of PIKFYVE in cellular pathologies, such as neurodegenerative diseases and metabolic disorders, by directly comparing the knockout phenotype with its wild-type counterpart. By offering insights into the mechanisms of action, researchers can better understand disease etiology and identify potential therapeutic targets.

In research and clinical settings, PIKFYVE Gene Knockout Cell Lines serve as essential tools for drug discovery, toxicology studies, and gene function analysis. Their availability supports investigations into novel therapeutic approaches to diseases influenced by phosphoinositide signaling abnormalities. Compared to other knockout alternatives, these cell lines are characterized by high specificity, reproducibility, and the feasibility for large-scale screening applications, thus providing a significant advantage in experimental design.

The value of PIKFYVE Gene Knockout Cell Lines lies in their ability to enhance the understanding of complex biological systems and inform translational research efforts. When utilized by researchers and clinicians, these models can accelerate the discovery of innovative solutions to pressing health issues, from cancer to metabolic dysregulation.

Our company stands at the forefront of advancing gene editing technologies, with a commitment to providing high-quality, customizable biological products, including the PIKFYVE Gene Knockout Cell Lines, thereby empowering researchers to push the boundaries of scientific discovery and innovation.
