LAT Knockout Cell Lines

Gene: LAT

Official Full Name: linker for activation of T cellsprovided by HGNC

Gene Summary: The protein encoded by this gene is phosphorylated by ZAP-70/Syk protein tyrosine kinases following activation of the T-cell antigen receptor (TCR) signal transduction pathway. This transmembrane protein localizes to lipid rafts and acts as a docking site for SH2 domain-containing proteins. Upon phosphorylation, this protein recruits multiple adaptor proteins and downstream signaling molecules into multimolecular signaling complexes located near the site of TCR engagement. Alternative splicing results in multiple transcript variants encoding different isoforms. [provided by RefSeq, Jul 2008]

Background

LAT Gene Knockout Cell Lines are genetically engineered cell lines that have had the LAT gene (Linker for Activation of T-cells) disrupted or deleted using targeted genome editing techniques such as CRISPR-Cas9 or TALENs. These cell lines serve as vital tools for studying T-cell signaling pathways, immune response mechanisms, and potential therapeutic targets for autoimmune diseases and cancer immunotherapy.

The primary function of LAT knockout cell lines is to investigate the role of the LAT protein in T-cell activation and downstream signaling cascades. By creating these knockout models, researchers can elucidate the specific contributions of LAT to T-cell development, function, and proliferation. Disruption of the LAT gene provides a clear demonstration of the effects on cell signaling, allowing for detailed analysis of the pathways integral to immune responses.

In the scientific landscape, LAT Gene Knockout Cell Lines hold significant value for both basic and translational research. They can be employed to assess the efficacy of new drugs and therapeutic strategies aimed at modulating immune responses, especially in the context of cancer and infectious diseases. Furthermore, these models can facilitate the screening of small molecules or biologics that target T-cell signaling, enhancing our understanding of immune regulation and paving the way for novel interventions.

One of the primary advantages of LAT Gene Knockout Cell Lines is their specificity and precision in modeling the absence of a key signaling component. Unlike alternative approaches, such as pharmacological inhibition, gene knockout provides a definitive insight into the effects of LAT loss in cellular systems without the ambiguity introduced by off-target effects. These cell lines are also easy to handle and can be customized to fit the specific needs of research protocols, making them versatile tools in any laboratory.

The value of LAT Gene Knockout Cell Lines lies not only in their application to cutting-edge research but also in their potential to contribute to the development of innovative clinical solutions. Researchers and clinicians using these tools can gain deeper insights into immune pathologies, thus fostering the development of targeted therapies and improving patient outcomes.
