Gene: ATF4
Official Full Name: activating transcription factor 4provided by HGNC
Gene Summary: This gene encodes a transcription factor that was originally identified as a widely expressed mammalian DNA binding protein that could bind a tax-responsive enhancer element in the LTR of HTLV-1. The encoded protein was also isolated and characterized as the cAMP-response element binding protein 2 (CREB-2). The protein encoded by this gene belongs to a family of DNA-binding proteins that includes the AP-1 family of transcription factors, cAMP-response element binding proteins (CREBs) and CREB-like proteins. These transcription factors share a leucine zipper region that is involved in protein-protein interactions, located C-terminal to a stretch of basic amino acids that functions as a DNA binding domain. Two alternative transcripts encoding the same protein have been described. Two pseudogenes are located on the X chromosome at q28 in a region containing a large inverted duplication. ATF4 Gene Knockout Cell Lines are engineered cellular models in which the activating transcription factor 4 (ATF4) gene has been intentionally disrupted. This genetic modification allows for the detailed study of the role of ATF4 in various biological processes, particularly those related to stress response, metabolic regulation, and cellular survival. By creating these knockout cell lines, researchers can investigate the underlying molecular mechanisms that govern these crucial physiological pathways, providing insights into how dysregulation of ATF4 may contribute to diseases such as cancer, neurodegenerative disorders, and metabolic syndromes.
The functionality of ATF4 gene knockout cell lines hinges on their ability to show altered responses to environmental stressors, such as oxidative stress or amino acid deprivation. Mechanistically, ATF4 is a key factor in the integrated stress response (ISR), modulating gene expression in response to cellular stress. As a result, when ATF4 is knocked out, downstream signaling pathways are significantly affected, enabling researchers to dissect the contributions of ATF4 to overall cellular health and stress adaptation.
In the realm of scientific research, the ATF4 gene knockout cell lines are invaluable tools for understanding the complexities of cellular stress responses and their implications in various diseases. They serve as a platform for drug discovery, enabling the screening of compounds that can potentially restore normal ATF4 function or counteract the effects of its deficiency. Furthermore, these cell lines hold promise in the development of targeted therapies, particularly in cancers where ATF4 modulation plays a critical role in tumor growth and survival.
Compared to alternatives, such as wild-type or overexpressed ATF4 models, the ATF4 knockout cell lines provide a clean background for studying the loss of function without the confounding effects that overexpression can introduce. Their specificity enables more accurate results and helps contextualize findings within the broader scope of stress-related pathophysiology.
For researchers and clinicians focused on understanding the intricacies of cellular responses and their implications for health and disease, ATF4 gene knockout cell lines represent a compelling opportunity. They empower users to explore novel therapeutic approaches and advance knowledge in cellular biology.
