ACTN2 Knockout Cell Lines

Gene: ACTN2

Official Full Name: actinin alpha 2provided by HGNC

Gene Summary: Alpha actinins belong to the spectrin gene superfamily which represents a diverse group of cytoskeletal proteins, including the alpha and beta spectrins and dystrophins. Alpha actinin is an actin-binding protein with multiple roles in different cell types. In nonmuscle cells, the cytoskeletal isoform is found along microfilament bundles and adherens-type junctions, where it is involved in binding actin to the membrane. In contrast, skeletal, cardiac, and smooth muscle isoforms are localized to the Z-disc and analogous dense bodies, where they help anchor the myofibrillar actin filaments. This gene encodes a muscle-specific, alpha actinin isoform that is expressed in both skeletal and cardiac muscles. Several transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, May 2013]

Background

ACTN2 Gene Knockout Cell Lines represent a significant advancement in the field of molecular biology, specifically designed for the functional analysis of the ACTN2 gene, which encodes the alpha-actinin-2 protein vital for muscle structure and function. This product is engineered using precise genome-editing technologies, such as CRISPR-Cas9, to create a cellular model wherein the ACTN2 gene is entirely disrupted. As a result, researchers can effectively study the consequences of ACTN2 deficiency, shedding light on its roles in myopathies and cardiomyopathies associated with mutations or deletions in this gene.

The key functions of these knockout cell lines lie in their ability to unveil the mechanistic pathways involved in cellular responses tied to the ACTN2 phenotype. By utilizing these cell lines, researchers can observe alterations in cellular morphology, contractility, and signaling cascades, providing insights into muscle development and disease mechanisms. Furthermore, the ACTN2 gene knockout models are invaluable in high-throughput screening applications, allowing for the identification of potential therapeutic targets and the testing of drug candidates aimed at reversing or mitigating the effects of muscle-related disorders.

Scientifically, these cell lines hold exceptional importance in both research and clinical settings. They serve as a critical tool for elucidating the pathophysiology of diseases linked to actin filaments, offering a platform for understanding disease mechanisms and accelerating drug discovery processes. Compared to conventional models, the ACTN2 gene knockout cell lines present distinct advantages, including greater relevance to human physiology and the potential for more accurate preclinical assessments.

For researchers and clinicians invested in muscle biology or genetics, this product provides a unique opportunity to enhance their studies with a model that accurately reflects human conditions associated with the ACTN2 gene.
