ABCB4 Knockout Cell Lines

Gene: ABCB4

Official Full Name: ATP binding cassette subfamily B member 4provided by HGNC

Gene Summary: The membrane-associated protein encoded by this gene is a member of the superfamily of ATP-binding cassette (ABC) transporters. ABC proteins transport various molecules across extra- and intra-cellular membranes. ABC genes are divided into seven distinct subfamilies (ABC1, MDR/TAP, MRP, ALD, OABP, GCN20, White). This protein is a member of the MDR/TAP subfamily. Members of the MDR/TAP subfamily are involved in multidrug resistance as well as antigen presentation. This gene encodes a full transporter and member of the p-glycoprotein family of membrane proteins with phosphatidylcholine as its substrate. The function of this protein has not yet been determined; however, it may involve transport of phospholipids from liver hepatocytes into bile. Alternative splicing of this gene results in several products of undetermined function. [provided by RefSeq, Jul 2008]

Background

ABCB4 Gene Knockout Cell Lines are genetically modified cell lines designed to specifically disrupt the function of the ATP-binding cassette sub-family B member 4 (ABCB4) gene, a critical player in lipid transport and cellular homeostasis. The knockout of this gene leads to alterations in the lipid composition of cell membranes, thereby providing researchers with a powerful tool to study the functional implications of ABCB4 deficiency.

These cell lines operate through a homologous recombination strategy that effectively eliminates the expression of the ABCB4 protein, allowing researchers to investigate downstream signaling pathways and physiological consequences associated with diminished ABCB4 function. In particular, the ABCB4 protein is known for its role in the secretion of phospholipids from hepatocytes into bile, making these knockout cell lines invaluable for studying liver physiology and cholesterol metabolism, as well as drug-induced liver injury models.

The scientific importance of ABCB4 Gene Knockout Cell Lines extends to various research and clinical applications, particularly in hepatology and pharmacology. They are essential for elucidating the mechanisms behind liver diseases, drug toxicology, and lipid-related disorders. By utilizing these cell lines, researchers can gain novel insights into the pathophysiology of diseases linked with ABCB4 dysfunction, as well as explore potential therapeutic strategies targeting this crucial regulator.
